My colleague would like to stain Branchiostoma using Fuchsine basic. Does anyone have experience with this technique? We're looking for details on concentrations since we can't really find anything useful on the internet. Everyone seems to use it as an additional staining agent, but not as sole agent. I'd be grateful for any help you can give us!
Procedure
1. Immerse slides in Basic Fuchsin at 65°C.
The time required varies depending on the thickness and type of embedding media.
2. Rinse in 2 changes of distilled water.
3. Stain in Basic Fuchsin at room temperature for 20 minutes.
The time required varies depending on the thickness and type of embedding media.
4. Rinse thoroughly in distilled water.
5. Dry stained sections on a 40°C hot plate.
6. Mount sections.
Stain Results:
Nuclei - blue
Cytoplasm, Mitochondria, Cilia - reds or pinks
Cartilage / Cell granules - blue to purple

Hi Sandra, basic fuchsin (or pararosanilin) is part of the Schiff's Reagens used in PAS. It is also part of the carbol-fuchsin solution of Ziehl-Neelsen. As counterstain it is used for example with Gram's stain (0,1% basic fuchsin).
I think as counterstain it is an "over-all" stain. Do your colleagues want to highlight a special detail in the fish?

Basic fuchsin staining is sensitiv to rinsing with diluted alcohols. After staining rinse with water, let the slide dry for half a minute, then quick rinsing in absolute ethanol and xylen (or similar).
