I'm currently writing my thesis on the MALDI-TOF-MS of apple condensed tannins, and aimed to purify plant extract via Sephadex LH-20, using a wash of 4x 90:10 methanol/water, then 4x 70:30 acetone/water. I noticed a significant removal of sugars, as expected, however signals for galloylated condensed tannins were noticeably increased relative to the ungalloylated condensed tannins. I am struggling to rationalise this observation, could anyone provide any insight into the mechanism of separation via Sephadex LH-20?
