I managed to stop my PCR run after only 30 min at 16 C incubation. I found it out after 30 min and started the incubation again (16 C for 1h and 72 C 20 min) Do you think I can still use my samples? Thanks!
Yes your internal control samples should indicate if anything has gone wrong and pcr reagents are very stable ( they spend a long time at 95c) so an extra 30 mins at 16 and a short while at room temperature should not be a serious problem to the pcr working
Sorry, the components of the synthesis mix are lyophilized so they are not known to me. I decided to continue with the experiment so I will see tomorrow how the preliminar results will look like
