to see the microbial degradation it is necessary to sterilize the crude oil and as it is heat labile it is not a proper way to sterilize and also filter sterilization adds limitations due to its particle size. So which will be the best way to achieve the same ?
- Heat the crude, 180 C, for 2 h, in a closed stainless container which prevents loss of volatile fractions. Cool down overnight, open and handle under sterile .
You could mix in a water based biocide and then let the water settle out. This could remove some water soluble species, such as acetic acid from the oil and could also change the presence of any emulsions (suspended water in the oil). But it would not change vast majority of the hydrocarbon species present in the oil.

Mr Didyk how about setting those temp. at said time in autoclave itself instead of heating directly and what quality of stainless it should be for direct heating?
Mr. Smith how much time and in what concentration will you specify to treat the oil by given biocide? OR it differs from product to product?
The concentration that you treat with biocide would depend upon the chemical used. I think that rather than worrying about the concentration of the biocde (since many of them have poor solubility in oil), the bigger questions are how much water solution you would need to use to get an effective kill and how much biocide ends up gets picked up by the hydrocarbon phase.
So Mr Smith after trying different combinations of water and biocide we can achieve this but at what extent we will sure about the process? is there any way to determine that oil is 100% sterilized after the treatment?
The only way to know for certain would either be to grossly over treat or to run cultures of the oil. Please understand that there is no such thing as an absolute 100% kill, either using chemicals or by autoclaving. Bacteria can go into spore form when stressed, in which case even hard radiation will not kill them.

Ultraviolet irradiation may be a choice.
The heating temperature is generally not recommended for more than 180℃, in order to avoid coking or decomposition.
The introduction of functional chemical products, should pay attention to ensure removal.
Any stainless steel will do.
Heat in a closed SS container, the temperature Is the temperature of the bath. Sterilize small ammounts of the oil in sealed glass tubes so you will have homogéneos, sterilyzed crude oil samples ready available
