I have encountered similar issues, and here are a few things that helped: use less medium so the LNPs stay concentrated at the surface, don’t rush to change it out, and try rocking the plate gently. Split dosing (two smaller doses) can sometimes boost uptake too. Serum vs no serum is worth testing, since ApoE can help LNP entry.
