You have suggested that 1gm in 10 ml gives 10% solution is understood. But 5gm in 100ml will give 5% not 50% (w/v). So 2.5 gm in 100 ml will give 2.5 %.(w/v)
I've made 30% solutions (w/v; 30g / 100mL) as that was a good density for my 5x agarose gel loading dye. The people that are googling solubility and saying that it is 10% are incorrect. Solutions up to about 50% can be made (50 grams / 100 mL). When dealing with solutions where a significant fraction is your solute, one can't ignore the volume of your solute. Don't just add 50 grams to 100 mL of buffer; begin adding it slowly to a smaller volume and when the Ficoll is in solution, bring the volume up to the desired final volume. For preparation of density gradients, accuracy here is critical.
