I am running a size exclusion HPLC for protein samples in Waters Alliance 2695. My mobile phase is sodium phosphate buffer and the column pressure is within the recommended limit. When I start the batch, it proceeds nomally to 1--3 samples and aborts suddenly with the attached error masage. Please help with this.
It looks like there is a failure when transmitting the data between the HPLC and the I/O board on your console. I would try unplugging the serial cables, blowing both ends with compressed air (the male/wire end of the input as well as the female/computer port side) for both the instrument as well as the console. Inspect to make sure there are no bent or missing pings, reattach carefully and once the pins are aligned then firmly push them into the ports ensuring they snap into place (or screw into place).
This can also be a driver error so pull up the device manager and make sure that the driver is working properly (Start -> Right Click on Computer -> Manage -> Click Device Manager). If it looks good there then shut the system and the console down, let it sit for 30-60 seconds, and then boot them back up. Turn on the deuterium lamp and wait until it warms up and then run a system diagnostic on the instrument from the control software.
Are you actually using serial connections or USB? We ran into issues before with USB cables of low quality or high length. It also showed up as devices not being remote because the connection was lost.

Is your Chromeleon software updated? Has your configuration changed? The acquisition error makes it look like a communications issue but it could also be a hardware fault with your detector. The 'Malformed UX response' could be a data type mismatch, do you have backup configs?
Vincent Paul Schmitz I am using Chromeleon 6.8 but I am not sure if it is up-to-date. I am new in this field so I don't know how to troubleshoot this error. Do you know where to look for the backup configs?
I have never used Chromeleon, although I know people who have and they tell me it is terrible. There are tons of processes and subprocesses that all have to be running in tandem, if any of them are shutdown prematurely or out of order the software can seize in the middle of a MS SQL write, which would corrupt the table and necessitate a repair. This type of thing should be handled by your IT department, I can tell you some things that I would do but I don't know your situation pertaining to data storage, backups, or access to software authorization keys.

I don't know if the procedure is different in Chrom 6.8 (except that 'backup' is called 'export' in your version) but apparently this is how you backup data in Chrom 7:
Close all Chromeleon applications (Console/Studio, Instrument Configuration Manager, Administration Console, Services Manager).
Stop all Chromeleon services and the SQL Server service, in the following order:
Chromeleon 7 Instrument Controller Service; this stops the Chromeleon 7 Real Time Kernel Service automatically.
Chromeleon 7 License Service; this stops the Chromeleon 7 Discovery Service, Data Vault Service, Scheduler Service, and User Management Service automatically.
SQL Server (SQLEXPRESS) service.
Use backup software to back up the Chromeleon data files. Depending on the installed Windows operating system, the following folder has to be backed up:
Windows XP
C:\Documents and Settings\All Users\Application Data\Dionex\Chromeleon\
Windows Vista/Windows 7
C:\ProgramData\Dionex\Chromeleon\
The folder contains all important Chromeleon files, including local data vaults (when created in the default location), license file, instrument configuration, custom ePanels, etc.
The most important sub-folders and files stored in the default location are as follows:
DataVaults: Folder for storing the Standard (local) Data Vault(s)
ePanels: Folder containing the Custom Panels
InstrumentControllerConfiguration.cmic: Instrument Configuration file
Chromeleon7_xxxxx.cmlic: License file
UserDb.exml: User Database
GlobalPolicies.xml: Global policies settings file
After finishing the backup, start the services in the opposite order.
If I were you I would search through the support ("help") documentation within the software and call the software vendor to see if they can walk you through troubleshooting before trying anything else. A system restore might revert your settings if you're on Windows, however that could delete data if you're not careful and don't have everything backed up. If you can figure out how to back everything up AND you have the authorization key to then you can try to uninstall and reinstall the Chromeleon software, it's dependencies, and drivers.
