I have a problem with my fluorescence pH probe.
This probe has two different fluorescence emissions in PBS (pH 6 green emission, pH 9 red emission)
When I did the cellular image with HeLa cell (pH 6 and pH 9 with PBS or BTB solution), the probe did not show red emission at pH 9.
It gives only small difference in its fluorescence emission.
How can I control the cellular condition to solve this problem.

You mean that you have put leaving cells in two buffers at pH 6 and 9 and see no difference in the intra cellular pH. This is expected, the cells are alive and manage to maintain a physiological inside them.
