I already tried using DMSO but it was precipitated (turbid) in to the aqueous phase
My drug is oil and in aldose reductase assay the solvent system is aqueous so i used DMSO as solvent for oil but when i add that into the aqueous solvent system the oil is re-precipitated.
You can also try using a detergent. Non-ionic detergents such as Triton X-100, Tween-20, Brij-35, n-octylglucoside, and dodecylmaltoside are usually compatible with enzyme assays.
Well that means that your concentration of oil in DMSO if high! That is pointless to test if the oil when mixed with water is precipitating. Try lowering the concentration of oil in DMSO and add much less sample (eg. 10uL of oil in DMSO in 200ul of enzyme mixture, or even less sample if you can). Also try other solvents but I don't think that the result will be better.
