I modified mineral material with dibenzocyclooctyne- amine (DBCO). Ratio of DBCO to mineral material is 1:100. I could not detect DBCO by FTIRT, raman and UV. mineral material can not be solved for NMR. Solid NMR not available for me.
Is the reason for not identifying a DBCO a small amount of it?

you can try derivatization of the amino group and extend the length of the conjugation, such that lambdamax undergoes a red shift, you can also try isotope labeling enrichment of the amine nitrogen by N15 and then run NMR of this nucleus. An alternate approach can be to try convert the substrate into a fluorophore, then you can analyse using fluorescence spectroscopy also, which is known to be sensitive even at very low concentrations.

Or crush the composite (very fine powder), perform soxhlet extraction.
