I have been using Xylan from Corn core as a substrate to analyze xylanase activity. But surprisingly the control is also showing activities! I tried to purify it by dissolving in the NaoH and washing with 70% ethanol. The amount of precipitate substrate is very less and still it shows activities after the assay. Which means it defiantly detecting xylose!
Anyone please have another protocol of purifying xylan substrate? Or perhaps a suggestion of a company as replacement to Sigma?
Since xylan is a polymer and xylose is a small molecule, you should be able to rid the xylan of xylose by a size-based separation. There are at least 3 easy ways to do this: dialysis, gel filtration (also called size exclusion) chromatography, and diafiltration/ultrafiltration.

I'm not sure that you can; the substrate that you are using is not pure, you need PURE xylan to perform the experiment,
I faced the same problem and I had to completely change the substrate
