If you contact water with a gas at a certain temperature and (partial) pressure, the concentration of the gas in the water will reach an equilibrium ('saturation') according to Henry's law.

This means: if you increase the pressure (e.g. by keeping the vial closed), the CO2 concentration will increase. So it simply depends what concentration you need for your assay: 'CO2-saturated' water at low pressure or 'CO2-saturated' water at high pressure ... I would think ambient pressure is fine. Furthermore, if you saturate it at low temperature, you should keep it at low temperature afterwards or the CO2 concentration will drop.

(be careful with high pressures: most vials won't withstand much pressure)

Still I have one more doubt; Incase if we apply any pressure like high/low/ambient, how can find that the water got saturated with CO2. Can you tell me, will it be there any indication.

I think 30 minutes of bubbling should be enough to reach equilibrium. If you just apply a pressure, but there is no continuous gas flow, it will take much longer.

There are instruments to measure CO2 concentration in water, or maybe you could simply measure pH and see when you get a stable value.

CO2 saturated water is prepared by mixing CO2 and water at high pressure and room temperature for a long time to insure a complete equilibration of CO2 and brine. A second pump (pump A, Teledyne Isco Model 450D) is filled with the mixture and injects CO2-saturated water in the core sample.
