the plant been extracted using methanol solvent by soxhlet apparatus, the extract is water insoluble, I can not dissolve and diluted by water. so what's the ideal methods for performing MIC tests?
If some material is water insoluble than solubilize it in inorganic solvent w/v, and use it in tests, now you have to keep two positive controls, one as plain control or simple one, another control is treatment with solvent.
use the same extarction sovent. Tween could be also used and control these solvent during MIC test

You can try solubilization in propylene glycol/PG or dimethyl sulfoxide/DMSO solvents.
