We have an isolated tissue bath, panlab 4 channel connected to powerlab (Ad Instruments). We all accessory for tissue bath just we didn't have 95%O2 and 5%CO2.
Use a metallic pressure bottle connected with oxygen and carbon dioxide cylinders through proper regulators .adjust gas flow in pressure vessel accordingly to your requirement.. Out let valve of pressure vessel can be used as per your requirement
you can buy this mixture from comercial gas providers (people who comercially provide O2, CO2 and N2O gases for hospital use. They usually provide 95% O2/5% CO2 mixtures as well. Tell them you need this for Research and they should be exact in 5% CO2 (O2% is not so important).
You better use Carbogen gas provided by commercial gas companies with the same component as you need (O2 and CO2, 95+5%, respectively) metal gas cylinders. Mixing them in Lab is difficult, will not be accurate and needs more safety considerations.
Because in our country we are firstly used isolated tissue bath for the first time, in iraq there are no carbogen gas (O2 and CO2, 95+5%), therefore i ask question in RG
is there any solution rather than carbogen like mixer or others!!!!!!!
Until obtaining carbogen you could try an small air pump (like one used in aquariums) to aerate air to the physiological solution. It depends on the tissue being used and will work in some tissues.
surely you can find CO2 and O2 cylinders in hospital. using gas meter (like those in homes for gas counter) add about 6 Liters of CO2 (normally CO2 is not pure in commercial cylinders) and 95 L O2 in a 150L capacity plastic bag (Douglas bag), mix and aerate to your solution with small air pump as Dr Babaei said. If the pH of organ bath is around 7.3-7.5 hopefully it would work OK.
You could also produce the oxygen in an Buchner flask closed with a cork and connected with a hose from hydrogen peroxide using manganese oxide as a catalyst, then capturing the produced gas by leading the connected hose into a waterfilled vessel inverted in a waterbath where the gas displaces the water. Carbon dioxide is easily produced from carbonates and acid and captured by the same method, but this gas gets absorbed by water, so the volume can change if left too long above water. A vessel that is calibrated for the volumes to be added eg. the 95% O2 and 5% CO2 (easily done gravimetrically with water) can be used to mix the two gases. To produce larger quantities the above mentioned plastic bags seem useful. It is also worth trying to mix all the required ingredients to produce the desired mix of gases in one bag.
