How can I higlight the bacteria over the particles with Image J?

Hi,

this image represents a fouled RO membrane with bio- and colloidal fouling. i did try weka segemtation to auto detect bacteria and particles but without any succes. is there any suggestions how to do it ?

furthermore is it possible to analyse the particle size distribution ?
