I have data sample of insects of same taxonomic group and I need will estimate the richness species, e.g. ants group, crickets group. The estimate will be separated to group and each group, at first moment not will be compared between them.
The compared will be made with diversity species, using the Test t to variance.

You need to select the diversity and richness index for this. There are several indices. For more on this please read

Anyway, If you wanted to estimate species richness and diversity indices, you can use some free software PAST, Biodiversity pro, ESTIMATES, Community Ecology Package, OC-ORD and many more

I completely agreed with Gautam Sir, if you wish to work on species richness and abundance, you have to evaluate the Shannon and Weaver Index by using PAST or Biodiversity pro softwares which are freely available every where and easy to use.
I have a software (www.dives.ebras.bio.br), and I need complementary this index, therefore I need a index of the richness specie, easy.
Complicated and a very unethical, in my opinion, a negative vote, only because I explained my idea about a my software, what freeware and is used to many researchers in Brazil and South America.

I wonder if the negative vote is because your sentences are very difficult to understand: what is richness specie? Maybe you intend richness of species (or specie richness). Richness is simply the number of species (and you do not need index but only counts), while diversity comprises not only richness but also equitability
If you dowlnoad the programme from the PISCES website you will find that you have access to several methods of species richness estimation and you can copy over an excel spreadsheet to do this. Caution it is important to have a rational sampling strategy to be able to have confidence in the results
