I am studying landscape ecology of Indian grey wolf in Gujarat, where I will identify corridor using circuitscape software. I have study areas that cover 22000 sq.km, where I want to do transect based survey to detect sign, so I can use it for connectivity and also I want to conduct interview, so I can give specific value to each villages and that can act as cost in connectivity model. in my team I have one person and I have period of two year to collect data, as I am dividing study area into 15 sq.km grid, so I will have around 1500 grid, so it is becoming huge effort, so how I can reduce that ?
also I want to conduct interview and i am having villages more than 5000, so how I should select village.

You might want to consider random samples in case of large study area. Lay those grids in GIS software and generate random points using the command there so that the selection is not biased. You can also choose line transects that can connect through the peripheral points. Secondly, use raster based predictive habitat models to compare your sample results with that of overall area. Similarly, out of 5000 villages, select a smaller sample size of fewer villages to represent your data or maybe use volunteers for surveys, else hand out questionnaire sheets for indirect surveys. As you said I am doing the same, using random sampling to select grids from my study areas, and I will be using Moran's I test to eliminate spatial autocorrelation of presence record to use for habitat suitability and as well as habitat connectivity. for villages survey I will use volunteer as well as cluster methods.
