I read some many research paper about GC analysis but didn't get good result. i got same peak at to different time. I did not get result from bacterial cells. Con you suggest good procedure for GC analysis for both pure PHB and bacterial cell.
I disagree with Michael. I have so many papers and colleagues who have done PHBs nicely derivatized and analyzed by GC and GC-MS techniques. Just search good reputed publications and follow their protocols in ditto. Should not be a problem at all!

I agree with Michael! The ENTIRE PHB is not possible to be analyzed by GC! The monomers, otherwise, can be analyzed by GC after derivatization. However, the hydrolysis of PHB is often ineffective to produce monomers! I don't now about pyrolysis!
The question is: What information about PHB do you need?

I agree with Michael and Lauro. In terms of volatiles analysis of PHB some have hydrolyzed PHB and then methylated prior to GC.

I suppose one could more simply take the hydrolysis product to an HPLC experiment, or do SEC/GPC on the polymer?!?
Or one may also study it less quantitatively by pyrolysis/GC

Those who inject it directly in solvent to a GC inlet may just get a few pyrolysis fragments from the polymer injected into that highly heated zone.
