I would recommend the freely available FRAGSTATS software; it will allow you to do this very easily. The page also offers an R package for handling the output data if you so desire:

similar to Oliver, I suggest you the 'SDMTools' package.

Please, have a look also at the function 'ClassStat' as it provides the same metrics of Fragstat.
