We are trying to do RNA FISH on a human midbrain sample and are having an issue with auto-fluorescence from the neuromelanin naturally present in the sample.

Does anyone have a method to remove or clear neuromelanin from a sample without otherwise destroying the tissue?

I found that methanol and hexane can work in the literature, but I am concerned that this may destroy the silane layers the tissue is attached to, or otherwise compromise the RNA.

Any suggestions?
