I am preparing isothiocyantes (NCS) from corrosponding amine. I am getting new spot above amine on TLC. Can you please suggest any stain to ensure that product is NCS?
ITC isolate (dichloromethane (DCM) extracts) detection on TLC, silica gel 60 F254 plates (Merck, Germany): can be done after thiourea derivatization. In order to prepare thiourea, 0.5 ml of ethanol and 0.5 ml of amonia (25%) add to the concentrated DCM extracts. Keep the solutions kept in refrigerator for 12 h. Apply the thiourea derivatives on TLC (silica gel G60 F254) with the lower phase of solvent system of ethyl acetate:chloroform:H2O (40:30:30). After developing and drying, the plates spray with Grote's reagent which is adequate for identification of thiourea derivatives by blue color spots.
Preparation: Dissolve 0.5 g. of sodium nitroferricyanicle in 10 ml. of water, and add 0.5 g. of hydroxylamine hydrochloride and 1 g. of sodium bicarbonate. Wait until a gas is no longer evolved and then add 2 drops of bromine. Filter the dark solution and make up to 25 ml.
Procedure for Test: Dissolve a few mg. of the substance under investi-gation in a little water, and add an excess of sodium bicarbonate and 0.5 ml. of the reagent. If a sulfur compound is present, a purple-red color forms within 10 minutes.
