I came to understand that I am only doing a check of absorbance instead of internal calibration. Anyone with useful information should please help.

First you must prepare a series of standard solutions, determine the λmax, finally measure the absorbance of all standards at λmax, the measurements must satisfy Lambert - Beer's law, where the relationship between concentration and absorbance in direct proportionality, then the standard (calibration) curve must be a straight line.. from this we can conclude that the instrument works well without any deviation..

I agree with the posted answers and the answer of Mohamed ist very right.
Your question remains obscure and obsolete until you give us (the answerers) the name and typ of your spectrometer (single , dual or arrays).
The technical guide is ONLY VALUABLE if you have :
- a new or old spectrometer with new lamps.
- Have you the possibility to contact the maintenance of the spectrometer Cy (p.e. Perkin-Elmer) and ask to test or exchange the lamps. It is always expensive.
- If you use an old spectrometer with old lamps, you 'll obtain no valuable measures.
The answer of Qasim is only valuable wirh a good maintened spectrometer.
- You must be familiar with the cut off of cuvettes (quartz or plastic) and the cut off of solvents used (quality spectranal ) and not pro analysis.
-You must ill your cuvette at the right level with solvent.

Most of the Spectrophotometers have an in-built calibration mechanisms. If yours is different, why not contact the manufacturers if the manual did not give the procedure for calibrating the spectrophotometer. I just checked the Google and found the attached document. I have not read it, but see if it will help.
