I do not understand what you are about to do. I assume that you have an "elastic response" and want to extract elastic constants. If so, you will need at least two responses, say stress-axial strain and volume change- shear strain. In such cases, if the material is absolutely linear, elastic and isotropic, you will be able to find the constant by a system of two equations for two (corresponding) unknowns.
