I observe partial degradation of polar compound by water during RP-HPLC analysis. Does anyone have suggestions for RP-HPLC methods were this can be avoided. HILIC and NP have been examined. The compound is only stable in pH range 6-8.
25oC. Higher temperatures results in increased degradation. Lower temperature is not possible since our collaborators does not have possibility to cool their systems.
The conditions you listed that would minimize the degradation of your compound, suggest the use of HILIC conditions, eg. low water, high organic at a pH of 6 - 8. You mentioned that you tested HILIC - how did the compound react under these conditions?
