I am trying to perform adoptive transfer in EAE models using MOG(35-55) in B6 mice, but always got many dead cells after 3 days of restimulation invitro. For example, I start culturing 5x10^6 lymph node cells and splenocyte in 1ml RPMI medium (24 wells) containing 50ug/ml MOG. After 3 days, when counting cells with trypan blue, I found that most of the cells had been damaged, and when using the lympholyte M to separate dead-viable cells I only got 10^5 living cell in which CD4+ T cells only account for 20-30%. This mean that I have to use a lot of cells and peptide If I want to inject 5x10^6  CD4+T cell into the recipient. So can anyone give me a good suggestion.
