I treated cultured cells with Colchicine (0.5ug/mL) overnight, isolated the cells, and treated them with hypotonic solution (0.075M KCl). After stopping the reaction with Carnoy's fixative solution, I observed the chromosomes under a microscope, but only a few cells had visible chromosomes. Is there a way to improve the isolation and observation of chromosomes using a microscope?
