I want to learn about how to culture and increase the yield of mouse naive T helper cells isolated from the spleen of Balb/c mice. I also want to know about the doubling time?
The cheapest way to do this is using the lectin Con A. It will nonspecifically activate the cells in large cultures like t flasks in 3-4 days. Normally 1 spleen into 6 flasks.
it is similar to staphylococcus super antigens, it promotes Th1 activation mostly. if you need to look at Th1 and Th2 responsiveness to drug Tx, you need to use plate-bound anti-cd3 and anti-cd28 mAbs.
