I have filled a Balloon with nitrogen gas and attached it to a test tube and place it in a sample cube of Princeten Fergie Raman Spectrometer.Liquids and solids are easily detected but i have tried several times but gas peaks are not detected. For gas i always get florescence signal from test tube and peaks obtained from system only
Try to make a raman spectrum of water, and then pass the nitrogen through the water so that it dissolves in it. Take the spectrum again and find an extra peak of nitrogen. If it fails, dissolve (solubilize) the nitrogen in the aqueous solutions of any surfactant. Raman spectrum do the same as for water.
You could also try adsorbing the gas on to a solid substrate like activated carbon, take the Raman spectra before and after adsorption and compare the differences.
While your balloon has nitrogen content of 100%, the air in your lab has nitrogen content of about 78%, so the balloon isn't much of a help. In practice it is enough to focus light just into air and both nitrogen (~2400/cm) and oxygen (~1600/cm) peaks are visible. They may be somewhat weak, so high NA objective, significant laser power and moderate accumulation are recommended.
It means high Numerical Aperture, a constant for each objective. Usually, this number is written on the objective, something between 0 an 1, the higher , the better. As a rule of thumb, the higher the magnification of the objective, the higher NA.
NA is connected with effectivity of focusing and (back)collecting light, so the higher value, the shorter accumulation you will need.
