I am applying fem in my studies. I did all the variational formulation of the problem (Navier stokes equations 2D), but now i do not know how to make the global stiffness matrix for four points quadrilateral elements and six points for triangular elements?.

Your question is not very clear. I am assuming that you want to combine a linear rectangular element with a quadratic triangular element. If this is the case you will need a transition rectangular element with three points on the side connecting to triangle. Better yet, why can't you use a quadratic rectangular element?
