I have isolated metagenomic DNA from1 gram of soil and dissolved in 200 mcl water. I took 100 mcl from it and added 2900 mcl water. Then I have transfered in cuvette and took od it gave me 0.431. It translates to 21.55 mcg/ml DNA, Do i need to multiply by 3 to calculate the amount of DNA in 100 mcl, since the assay volume is 3 ml ? and then multiply by 2 to calculate per gram of soil ? This may be a silly question. help me. Thanks
