I am trying hard to isolated mitochondrial DNA from Onion leaves? Is it possible to isolated DNA or do i have to choose some other part as i could get to know that leaves contain high amount of chloroplasts? tried many protocols like differential centrifugation, sucrose gradient centrifugation but very difficult to get any mt DNA? can any one plzzzzzzzz suggest a protocol or what all things i should take into consideration????
thanks a ton Mr.Vasanth... well i have tried all the three protocols but couldnt get any DNA, infact getting good quantity of Genomic DNA...
yes there is a need to isolate the mitochondrial DNA..we need to sequence the mtochondrial genome
HI Artur... we have tried different homogenization procedures like liquid nitrogen, blending, etc. yes as you have suggested need to increase the starting material .. but can you please clear a thing to me.. is it ok if we select the leaves? i have gone through some literature which suggests that there is more number of chloroplasts in leaf than mitochondria?is it ok if we select the bulbs or scale leaves...? kindly suggest..
