I saw in some protocols where people prepare BSA-conjugated LPS, or A+ serum conjugated LPS in order to polarize THP-1 macrophages into M1. Do you know why? What is the benefit compared to LPS solutions prepared in medium or PBS?

M1 polarization of the Thp1 cells by LPS is signalled through TLR4 pathway. LPS conjugated to BSA has higher chance of binding to TLR4 receptor than free LPS because it is concentrated on the surface of BSA. Because of its higher molecular weight BSA has higher chance of coming close to TLR4 receptor bringing the LPS along with it. In this way BSA not only increases the efficacy of LPS with respect to TLR4 pathway activation, but few log fold lower concentration of conjugated LPS can do the same work as higher dose of free LPS.
Efficacy of LPS can be increased by conjugating it to other synthetic polymers such as PEG too with the similar mechanism. We conjugated host innate cationic peptides to carbon nano tube (CNT) and gold nano particles (GNP) to increase their efficacy to modulate host innate immune system. With the similar mechanism we increased the efficacy of Indolicidin by 1000 folds.
