Hi everyone!
Using CO antibody (Santa Cruz sc-33753), I couldn't detect the expected band at 40 kD, however, there is an obvious band appeared at 30 kD. I am not sure whether this band labels CO protein, as it is also detected in co-9 mutant. I also want to know whether co-9 is a null mutant and shouldn’t be detect the protein. I hope people who is good at CO protein detection could give me some suggestions, thanks!
Migration of proteins in Western Blot depends on many factors, including glycosylation phosphorylation etc.
There are cases that antibodies from different suppliers gives different band location. practically you can try also recommended positive control.

did you manage to detect the correct bands?
I also tried this AB and couldnt get anything around 40kDa. What I do get is a stained marker and 2 very weak bands around 25 and 35 kDa, probably unspecific
