I have been using the Immunotag Calmodulin Polyclonal Antibody ( Catalog number: ITT0612, Alexafluor 647) to label calmodulin in mouse brain tissues. I have been using it at the concentrations of 1:500 and 1:250, diluted in 1X HBSS, 0.1% saponin and 2% normal goat serum. I incubate the tissue with the antibody for 24 hours. However, I am unable to detect a good signal, and all I see is non-specific labeling. I tried looking at the G Biosciences site for literary reviews for the use of this antibody but was unable to find any. I was wondering if you would tell me the optimal conditions ( concentrations, buffers, incubation time etc) for the use of this antibody so that I can use it to get better labeling.
