Hi Bhargava Teja Sallapalli, YES you start the lysis step from the viral supernatant and continue with the following steps: Add 100µl of your viral supernatant to 300µl of RTL buffer, followed by adding 300µl of ethanol, transfer to columns, wash (RW1 & RPE), and elute. It works very well!

Hi, I would suggest to use Qiagen viral rna kit.

the yield of viral RNA will be very low, the viral rna kit adding the carrier RNA in it to increase the yield.

if you plan to use your kit, I think you could add some yeast tRNA (etc.) to the lyze buffer.
