I would like to conduct studies with mice using the Intraperitoneal route, however, nitidine chloride is indicated to dissolve in DMSO at 1mg/ml. if I need to administer a drug concentration of 10mg/kg of a 0.2 ml volume (dissolved in DMSO then PBS). the DMSO % would be over 20% which would kill the mice as the only recommended is 5%.
You final concentration of nitidine is only 10 mg/kg. If you, why not prepare it in the PBS directly, like 10- 20 mg/Kg or mg/L. It is a salt, which should have some solubility in aqueous solution. An analytical balance can weight 10 mg precisely without any problem.

Unfortunately, it forms a suspension if directly dissolved with an aqueous solution, making IP administration impossible. the available way around this is DMSO first then the aqueous solution. in this case the solubility is very low 1mg/ml. prev research have published that show they made stock of 10 mg/ml first in DMSO, i have tried the same 10mg/ml (10x higher than the rec) and it did not dissolve. only a 3mg/ml.
