Aloha all, we are having difficulty getting AHLs out of cultures grown in Petri dishes (various types of media, environmental microbes). The literature tends to report cultures grown in broth (liquid media), shaken, and then centrifuged to collect supernatant and pellet cells. As anyone has success with extraction of AHLs for Mass Spec analysis using cultures from Petri dishes? And if so, how much material did you collect to extract?
