I am trying to isolate CD163 positive population of horse tracheal samples. I tested the antibody of this kit separately using FACs and confirmed that it cross-reacts with equine macrophages from the same type of sample. That is why I bought the kit afterwards. However, I am using the MACs separator from Biotec and the LS columns from the same company (Biotec) and it doesnt seem to work. I checked at the Celltechgen LLC site and they dont seem to provide specific columns that would be compatible with that kit. They do provide only a stand.

Finally in their protocol they recommend using 100ul of dissociation buffer onto the column. However, that is far too low volume for an LS but even for an MS column. I am suspecting that these columns are not compatible or something is wrong with the protocol. So if anyone has used this kit before or has any advice would be much appreciated. I checked and there are no publications using that kit, yet.
