Worked brilliantly - 40uL sample (or standard), I found no need to dilute (was using lobster haemolymph serum), 80uL reagent and then 80uL 12N H2SO4 to stop rxn. Only thing I would say is always start at the same end with the multichannel so each column is getting the same incubation time.

I have used a similar kit for glucose and, as we usually have lots of samples, I always scale down the protocols for a plate reader. This should be completely feasible with your kit, and the few steps that you have explained are not a problem at all. In case you need some more help, just send me the protocol and I can give you hand.

No experience with this assay. Recently used Invitrogen Amplex Red fluorescence assay ( in 96 well plates). Very pleased with this one, less elaborate than what you describe

Thanks guys, think I will be giving it a go, turns out that the 30-60 sec intervals were just to give time for measuring, but if I use a multi-channel pipette, should be okay... now to work out whether 33% H2SO4 will be okay in a plastic 96 well plate... !

I have successfully used similar glucose kit (Glucose liquicolor; Human GmbH) on the scaled-down measurements on 96-well microplate. Measurement is done at OD 540 nm. With this kit adding acid to the samples is not needed. If the plate reader does not have the possibility of temperature control then incubation of the plate in usual termostate is sufficient.

Well we didn't have all the correct 'ingredients', and decided it would be more cost effective/less wasteful and get a kit (most of the reagents we would only use once)... Turning out to be more trouble than it's worth though, I will keep you posted!

I am used to measure glucose levels from plasma with Wako kit (439-90901) which we scaled down to be used in microplates. Recently, following references from colleagues, I also bought the same kit (Sigma GAGO-20) to try to reduce costs. I did not try yet to scaled down, but I would really appreciate if you could send me your protocol.

The constant use of kits has always worried me as I feared it lessoned a PhD students deeper understanding of the chemistry and invention . May you continue to adapt many more kits in your doctoral studies - it says a lot about your intellect, understanding and ability.

Your protocol sounds o.k. to me, we have done the assay similar as yours. No problem.
