I have never used this program. I normally use the DNASTARLasergene set of programs, specially the SeqBuilder one. I really love this one. You can easily make virtual construct and check in advance if all your prediction are right before (e.g. that everything is in the same reading frame or the translation changes when mutating specific nucleotides..) before starting with your cloning or site directed mutagenesis.
Seqbuilder costs like £500 for a single academic licence. Haven't used vectorbuilder before, but it looks like the company is selling the plasmids that you design from the building blocks that they have available (not sure if you can use it for sequences they don't have already available).
If you are looking fro a good free software for in silico cloning purposes, I recommend ApE plasmid editor http://biologylabs.utah.edu/jorgensen/wayned/ape/
If you are asking if anyone has experience ordering from the company behind it (Cyagen), then I've not had the opportunity but I have used similar companies before and it can be much more time and money efficient than doing it yourself.
Sorry was wanting to know if anyone has used this company to make a vector. Its through Cyagen. The design we have under control but wondered with complicated vectors, if it was cost effective to have someone else construct. Thanks for your input.

I used vector builder to design and order AAV particles last year. It was my first time to use AAV. The website and customer/technical support were both helpful in designing the vectors, which worked very well.
