You can follow the procedure of NEB endoproteinase aspN, link is below:

Prepare buffer 1X Endoproteinase AspN Reaction Buffer 50 mM Tris-HCl, 2.5 mM ZnSO4 (pH 8 @ 25°C) for reaction. Dissolve your Endoproteinase (Usage Concentration 100 ng/μl) and Substrate (varying) then incubate the reaction mix for some time for degradation.
