I have not use this NBT/BCIP, but I am surprise that you can not reuse your membrane : I have use the NBT from Sigma, and I can reuse the membrane. I reuse without striping, but it is a personal choice. I just do again blocking.

The limitation is the sensitivity of this method. it might be due to the inhibition of the enzymatic reaction by the colored product deposition at the antibody position.

Hello! Just to tell that I curently use NBT/BCIP to reveal blots. It works pretty well for me. And I can reuse them if they are not dried after the NBT/BCIP revelation (put them in de-stripping solution). After all the proper process I could revealed them again with Fast Red TR / Naphtol. But at the end I choose to use first Fast Red/Naphtol because it's red (scan it wet) and at the end use the NBT/BCIP revelation for the second protein. And finally it's nice you have one protein in red and the second in violet. When the membranes are dried yo can keep them for long time.

I am having a problem in my NC membrane blots. When I use ponecus dye my proteins bands are visible very nicely and equally but when I wash the Dye membrane  using  Grade-C water and then use NBT/BCIP to visualize my AP-linked 2nd, the background comes very quickly but my beta-actin band don't develop even I incubate overnight only faint band develops and I get alot of background. how to solve the problem?
