I have a problem while detecting pool of amino-acids. I am using Phenomenex detection kit and Zebron ZBAAA 10 m X 0.25 mm capillary GC column. I am taking all precautionary measures while preparing samples however, during injecting in GC-FID (Alignet technologies) i could hardly observe 3-6 peaks of amino acids instead of 20-21. I am using plant samples for amino acid detection. I am also observing giant peak at the starting which i presume is not amino acid peak. I observe the same peaks while using internal standard too. Please help me folks how can i overcome this trouble shoot. I am using He as a carrier gas.
Well, the firstt peak (giant as you said) observed is related with solvents. If you are using Phenomenex kit, either for free or hydrolyzed aminoacids, the first peak you will observe, using the method recommended by the manufacturer is the Alanine (ALA) at about 1.5 min. As you say, you only obserfve a few peaks in your chromatogram, but are those peaks from one of the calibrating samples? If this happens in one of your sample it isprobably due to the sample composition, but if it is related to one of the calibration samples supplied by the manufacturer, it is probably related with the sample preparation, mainly to the derivatizing reaction
