The main thing is to have a 50mM Co2+ solution and indeed if you prepare a 50mM CoCl2 solution from its hexahydrate powder it is 50mM in terms of cobalt content. Note that the associated anion doesn't really matter : you could use cobalt sulfate for instance.

Yes, you may use CoCl2.6H2O. You need to go by the MW of CoCl2.6H2O, and not by the MW of CoCl2.
