We used Thermo Scientific™ Varioskan™ Flash Multimode Reader for the fluorometric measurement. We load 60uL of sample into each well so that it covers the well. We devised the program such that it make measurements every 1 minutes, and in between there are shaking of plate in between. We took 46 measurements for a 45 minutes period.
However, there was a step called "Executing drift compensation" which messes up the data and increases or decreases the reading in a period of time. Why does it happen? And how could we solve the problem?
