I am using the Florisil column to elute α-farnesene and its oxidation product; conjugated trienes from apple peel extract with Hexane and hexane: diethyl ether mixtures.
However, I can't completely elute the volatile compounds (non-polar) from the extract using this particular column as well as solvent mixtures.
Here is the list of my questions:
1. Changing the absorbent will help to elute 100% compounds?
2. Changing the Hexane mixture will help for the elution? if so how do find the right mixture ratio?
Some of my questions are very basic, since, I am new to use the methodology to separate, identify and quantify matters.

Start with TLC (thin layer chromatography). Florisil TLC plates are available commercially. Use different ratios of hexane and ether; ether tends to make the compounds move more on the TLC plate and the column. You may find that you need a "stronger" solvent than ether such as ethyl acetate. you can look for the movement of the compounds after the TLC plate has run with iodine or a water spray.
Make sure you run your TLC plate properly- make sure the chamber is well sealed, don't overload the plate.
