I am attempting at enrich plasma exosomes total population with neuron-derived exosomes. In order to do this, I am gonna try a Direct immunoaffinity capture (DIC), using magnetic beads conjugated with my antibody of interest. Since I actually have no experience with this tecnhique with exosomes, I am not sure about the size of the magnetic beads to use. I think I will need high magnetic mobility. Do you have any suggestion about the size to choose?

Which magnetic bead to chose depends more on your downstream application. Whats even more critical than the bead size is the number of beads used per isolation. The number of beads used is also depending on the downstream application. For downstream flow analysis the fewer beads the better. this is to ensure sufficient number of exosomes to dock onto the solid surface of the magnetic beads and provide a strong signal as the bead:exo complex travels through the flow cell. For e.g. downstream western blotting the situation will be different. For efficient capture you will aim for a large total surface area = many beads. This will provide enough surface for exosome docking and detection by e.g WB. The 2.8 µm Dynabeads works well in both cases but the number of beads used per capture will be different for flow and WB.
