I just inoculated a new E.coli culture (k12 Gal+ strain) and I recognized something I can't explain, the bacteria lawn (by excessive water) has a branchy appearance, any explanations for this?? I haven't seen this in any bacteria strain I have had before. Is this typical for e.coli?
No there is no contam. I inoculated the culture freshly (about 20h ago) from a sterile culture tube. I make a phage test to check whether it is sterile or not
It might be due to the excessive moisture on the media surface. Anyway, the plate looks bad to be used for any good culturing method/technique. Try to culture on a fresh and dried plate and compare/evaluate the growth results.
For my purpose the plate is ok, it is just a subculture to check if there were any e.coli viable in the vial, because it was about 1 1/2 years old. Of course I will do a subculture and not proceed with this one. But in my opinion the plate doesn't look all that bad because I don't have any contaminations :)
I recommend reinoculating from the same plate to see if you reproduce the effect. It sounds like simple swarming and dry plates will fix that, as noted by others.
