My plasmodium culture has been contaminated for some time, but this time I have a new problem - I see some fibre like structures in the culture flasks and that fibre also now appears in stored Blood. I am confused about the fibres. Could these fibres be fungal or otherwise? I have some doubts on ACD, can somebody tell me if ACD is not working properly then is this a possible reason for these fibres?
If the fibres are found in blood to, then that might be cotton. If u can make a slide and view, that could clear all ur doubts. A simple Giemsa staining used for malaria parasitemia counts would do.
Quite likely to be fungal contamination - any culture with such contamination should be disinfected and discarded. If there's fungus in the culture, it's likely to be in the incubator too. Deep cleaning and disinfection with Virkon multiple times may be necessary to remove it. Place 2 open nutrient agar plates in incubators for 1 hour after cleaning, close then incubate at 37 and 25 degrees C to detect residual contamination. Pictures of contaminants can be seen at
