What is the exact process for capping of nanoparticles?normally the BSA is fixed using glutarldehyde or some other reagent . And in which case the sodium borohydride is used to reduce any schiffs base formed
In nanoparticle BSA cap the nano particle . The BSA is not get unfolded and it form like a matrix around nano particle. If the protein denatured or unfolded, you can examine them with fluorimeter or even with uv -vis spectroscopy. 280 nm absorption band shows the nature of protein. I have experience with BSA -Gold nano particle conjugates, the protein is stable for 2-4 hrs in conjugation with nano particles.
1.Effect of borohydride on BSA was linked to the impact of protein unfolding on the involvement of specific cysteine residues in intermolecular and intramolecular thiol-disulfide interchange and thiol oxidation reactions.
2.There are many factors of stabilization of disperse systems. Characteristic of proteins are two. The first structural-mechanical is kinetic. On the surface of the particle there are films that have elasticity and mechanical strength. Destruction of them requires a certain amount of energy and time. The second factor is electrostatic (thermodynamic). The appearance of an electrical potential on the interphase surface is caused by surface electrolytic dissociation of the carboxyl and amine groups of the protein.
